MPS 01-04 THE CARDAIC PROTECTIVE EFFECT OF POTATO PROTEIN HYDROLYSATE

Abstract
Objective:
In aging and obesity induced cardiac remodeling includes cardiac hypertrophy and cardiac fibrosis. Gelsolin (GSN), actin-binding protein, can regulate the cardiomyocytes morphology and hypertrophy and TGF-β can induce fibroblasts proliferation. But the relationship between TGF-β and GSN is still not well known.
Design and Method:
We evaluated the TGF-β and GSN expressions in a high fat diet (HFD) fed aging rat animal model and gave anti-hyperlipidemic drug probucol and heart protective functional food alcalase potato protein hydrolysate (APPH) as the treatments.
Results:
As the results shown, the APPH (45 and 75 mg/kg/day) and probucol (500 mg/kg/day) treatments can reduced the aging and obesity induced cardiac hypertrophy and fibrosis. Further, the GSN dependent hypertrophy signaling pathway was also be inhibited with an anti-TGF-β neutralizing antibody in HFD aging rat serum cultured H9c2 cells.
Conclusions:
In conclusion, TGF-β might be not only the key role of cardiac fibrosis but also involves the aging and HFD caused GSN dependent cardiac hypertrophy. TGF-β in serum could be a therapeutic target in attenuation of hypertension or pressure overload-induced cardiac hypertrophy and heart failure.
